Secretion of mCherry in wheat coleoptiles. Confocal images of wheat coleoptiles infected with PH-1 or the strains expressing either the non-secreted version of mCherry (PtrpC-mCherry-TtrpC) or the secreted version (PtrpC-SPosp24-mCherry-TtrpC). The strain expressing the non-secreted version displayed accumulation of the mCherry fluorescence signal solely inside the hyphae. Whereas the strain expressing the secreted version displayed accumulation of the fluorescence signal in the periphery mainly localised towards the tips. Graphs a, b, c and d indicate mCherry signal intensity determined along the diameter of the hyphae, taken at the positions indicated by yellow lines in each image enlarged and labelled with the same letter. The yellow arrows point to the selected hyphae used for this analysis. Wild type strain PH-1 was used to set confocal conditions. Confocal images were taken 2 dpi
